The researcher aims to develop a single bilayered hybrid construct able to promote in vivo, the simultaneous regeneration of both cartilage and subchondral bone involved in an osteochondral defect. Different types of culture medium, supplemented with biomolecules known to have an influence on stem cells differentiation, will be systematic attempted to obtain the ideal formulation for inducing osteogenesis and chondrogenesis in a bi-chamber bioreactor system. For osteogenic differentiation, supplements as vitamin D3 or dexamethasone will be tested. For chondrogenic differentiation, growth factors such as IGF-1, BMP-7, BMP-2, TGF-β1 and TGF-β3 will be evaluated. Cultures will be characterized regarding the relevant markers for each lineage.
